There's a LOT of changes to Knuckles' voice compared to the final. He has a gruffer, more stoic tone of voice in this demo. This does, however, cause him to lose a bit of testosterone in the final version. Some of his voice clips are either missing from or identical to the final game.

Behind Knuckles' Voice

Knuckles has a gruffer, more stoic tone of voice at this point. 95% of his lines would be redone by the same actor in time for the final. Two remnants of this early voice direction exists in the final game - the infamous "Oh no!" clip from when he takes damage in character battles is reused from his story's intro cutscene in this build, while the "Give it your best shot!" clip from when he's on his last health also in character battles is taken from when Knuckles is about to fight Chaos 2.

Compared to the final game, only a few of Knuckles' voice clips are missing.

Audio Differences

A majority of his audio are present on the game's disc. While most of his audio can't normally be heard in this demo, using the Tokyo Game Show menus will make it possible to hear most of his voice clips in-game, although not everything is present (such as his final "Ahhh! Eggman!" and "All right! Put 'em up!" audio clips).

His second audio contained a slightly different script. There was no "I", and "these" became "those" in the final. These are used in the Adventure Fields for Knuckles' story.

Unused Audio

Unused in this Build

While most of Knuckles' voice clips can be heard during Adventure mode (if one were to use the TGS menus), some go entirely unused due to the cutscenes either still using Japanese voice clips or having no voice clip present at all. For example, in Knuckles' story, a voice clip that plays before Knuckles starts an adventure to find the missing pieces of the Master Emerald is not used, as it doesn't play in this build, and two voice clips that play before fighting Chaos 2 both go unused here, since this demo still uses the Japanese voice clips there.

Entirely Unused

Out of every audio unused in this build, only five never saw any use in the final either, even after being rerecorded.

As seen in the "action stages" section above, there is a completely different voice clip of when Knuckles loses a life from getting hit by an enemy with 0 rings. Just like the final, he is silent in-game when dying, so this goes unused entirely. Additionally, there are menu voice clips for SEGA/Sonic Team logo, Adventure Mode, and Trial Mode that are unused in both this build and the final. Also see the "menu" section above.
